I have a #4 mk1with the 600/300 flip aperture sight. My front sight is marked 015 and has a total height of .320". What front sight do I need to be on the mark at 100yards?

I am not an Enfield expert!
 
However, with that said, you need to shoot your rifle with your existing front post sight and determine how high or low it is hitting with your existing .320" front sight.

I have some spare blades which may be closer than what you have installed now, but you need to calculate how much you want to change the point of impact.

iirc  a change in blade height will be about 2" at 100 yards

Bear in mind the 1.8" change applies to 174 Gr Mk7 ball only.
You'll need to shoot, change & measure with any round having a different trajectory.
The path should be about -9 1/2" @ 300 yds so you'll need to back calculate for 100.

When I read that link to zeroing above it said that the .075 was the largest blade and I wrote my last post requesting one. It seems using the formula that the .075 isn't tall enough anyway. Transposing the formula tells me I need a .120 using #7 ammo.
 
Now, on ebay, I found and purchased a +1.14 from Southwest Shooters Supply. I can file it to the correct size once I work up a load.

Bear in  mind those numbers are to accurately zero for 300 yds. Not to change a 3000 yd zero to a 100 yd one.
You can use them, but you have to figure out not just the zero error, but the change in zero as well.

You can file the sight blade to bring the POI UP, but the actual change in heights is the base, not the blade. All the blades for a set are the same height.

I have finally done a proper fix on this rifle. I just purchased a Singer rear sight on ebay. Now I won't have to cobble, file, or what ever to get it to sight where I want it.
 
I have noticed in the photos that these sights have two marks at each of the range numbers. Does this mean I must experiment between these marks to get the right adjustment?

Not sure what you mean about two marks Lily's dad. 
I Think you will find the upper mark is for 50 yards further, as in, 200, up to next mark = 250, then 300, etc.  One click is less adjustment than these 50 yd marks, so they can be got pretty well spot on.

Notice there is a scratch mark at the yard number and then a shorter one just above it.

Yep.
2 =200 yds
- = 250 yds
3 = 300 yds
- = 350 yds
